Din considers him a Mando, because he has Mando blood, where as Bo does not as just as that one man said, they don't see Fett as a Mando. She may hold that sentiment while those that Din were apart of and other fringe groups may consider them that. Din having basically been raised to be a bounty hunter once the Empire arose, see it differently. And that he is a clone, That other lady RUBS that one in HARD.
It was actually Bo-Katan who brought up him being a clone, and Boba kinda provoked her by scoffing at the idea of taking Mandalore back.

An unrelated observation: the fact that the Dark Troopers assaulting the bridge stopped and switched back to their guns when Luke landed on the ship a level below, as well as their generally perfect marching coordination, is a pretty strong indicator that the DTs are tactically networked with each other, unlike B1s which clearly have no awareness of what other B1s see. The only B1s that had that kind of coordination were the centrally controlled ones that were unloaded from the MTTs during Phantom Menace, and we never see that kind of coordination from independently operating B1s.
